Premier Anti-HCV ELISA Test is based on the principle of inter-second antibody enzyme immunoassay procedure. Wells of the microwell strips are coated with purified recombinant Hepatitis C Antigen (HCV). Serum, appropriate controls and monoclonal antibody of human IgG horseradish peroxidase conjugate (anti-Human-IgG: HRP) are incubated with the bound antigen. During the incubation period, Anti-HCV, if present, will bind to the solid phase antigen and simultaneously bind to the anti-IgG: HRP. Unbinded Sample and conjugate will then be removed by a wash step. Working chromogen solution is added to the wells and allowed to incubate. A blue or blue green colour develops in proportion to the amount of Anti-HCV present in the sample. The enzyme reaction is stopped by the addition of a Stop Solution which changes the blue-green colour to yellow. The optical absorbance is determined with a spectrophotometer set at 450 nm.
